Genome-Wide Rad-SEQ Reveals Adaptive Divergence Among Seven Stream Stoneflies Along a Nationwide Latitudinal Gradient in Japan

Abstract: We investigated genomic variation in stoneflies in relation to environmental conditions using a multi-species approach. These samples were collected from 23 sites distributed across four geographical regions of Japan spread along a nationwide latitudinal gradient. Double-digest restriction-associated DNA (ddRAD) libraries were constructed and statistical analyses were conducted to determine the effect of environmental variables on genomic variation indicative of environmental adaptation. Data showed significant correlation between genomic variation and two environmental variables, precipitation and altitude. Additionally, comparisons of genomic variation between species revealed the possibility of parallel evolution and high homology between the genomic sequences of stoneflies and carp. We demonstrated that using multiple-species approaches may be more productive in understanding the effect of environment on adaptive evolution.
